Procedure( Part One )
Slice kiwi fruit into 6 pieces per student
Add 20 ml of Suave Clarifying Shampoo. This Shampoo is essential to the extraction procedure.
Place in zip lock bag.
Place bag containing kiwi in another bag
Use a Falcon tube to mash your Kiwi Fruit.
Mash for 10 minutes
Make sure that the Kiwi is almost in a liquid form
Cool for 2 minutes
Heat for 2 minutes in the water bath at a temperature of 55oC
Spoon mixture onto a cheese cloth covering a beaker
Use the cheese cloth to strain the lysate or mixture of the Kiwi fruit.
Pour about 7 ml of lysate into a clean Falcon tube.
Place on ice for 5 minutes
Or store overnight
To extract the DNA Tilt the tube at a 45o angle and drip ice cold 95% ethanol down the sides of the tube until you have a layer of alcohol floating on the top of the lysate . Take special care not to mix.
You should see strands of DNA appear at the interface of the alcohol and the lysate.
